Experimental agents for breast cancer reduces bone metastases

Researchers have reduced inhibit metastasis of breast cancer to bone with a rock experimental agent to be a protein that was found over-expressed in breast cancer metastasis. In a study on mice, the team of researchers from Tufts University School of Medicine, the Sackler School of Graduate Biomedical Sciences at Tufts and Tufts Medical Center reports that the rock or inhibition of Rho-associated kinase in the early stages decreases in bone metastatic breast cancer by 77 percent and the overall incidence of metastases by 36 percent. The results suggest that the rock can reduce a target for new drug treatments for metastatic breast cancer. "Even if the disease causes serious and requires primary treatment, metastases represent over 90 percent of breast cancers linked to deaths. There is no treatment for the removal of the metastasis. Creating ROCK brings role in the spread of breast cancer and identification of agents inhibiting ROCK a step closer to an approach that could reduce metastasis in the future, "said lead author Michael Rosenblatt , MD, professor of physiology and medicine at Tufts University School of Medicine and a member of the program of cellular and molecular physiology at the Faculty Sackler School of Graduate Biomedical Sciences at Tufts. Rosenblatt is also dean of Tufts University School of Medicine.

"We also found that precipitate with shRNA - small RNA hairpin - ROCK expression slowed metastasis. Thus, cancer cells migrate to a global transportation equipment is necessary. ROCK directs the formation of this device, but using the ROCK inhibitor and shRNA that transport mechanism of cells to be ineffective in reducing significantly to metastasis of breast cancer to bone, "said First author Sijin Liu, Ph.D., research professor and a member of Rosenblatt's laboratory at Tufts.

"This study also showed that a specific RNA, cluster 17 is connected to 92, with the words ROCK and metastasis of breast cancer. The micro-RNA in response to inhibition of Rock, the information on the mechanism of metastasis and offers Voyage, a finding which is of particular interest to researchers on the role of RNA fragments in the expression of gene is, "said Liu.

To see Rosenblatt, Liu and colleagues used fluorescence imaging for Effects ROCK ON metastasis of breast cancer. Researchers found that causes the insertion of the skirt up with non-metastatic cancer cells, cells metastasize to multiple secondary sites, whereas cells without rock exposures remained localized. The researchers used an investigational agent (Y27632) or shRNA reduction of ROCK activity in seven of the mice with metastatic disease, finding a significant reduction in bone metastases compared to six untreated mice.

Breast cancer is the second most common fatal cancer in women, affecting nearly one eighth of women in the United States. Bone is the most common site of metastasis of breast cancer, which affected three times more often than the lungs or liver.
